- Mechanism of dysregulated expression of the pro-inflammatory cytokine and chemokine genes including interleukin-8 (IL-8)
- Post-transcriptional mechanisms that regulate genes characterized by the presence of AU-rich elements (AREs) in the 3'-UTR, key regulatory elements that accelerate mRNA degradation
- Recently, microRNAs (miRNAs) have been found to be key players in post-transcriptional mechanisms. The mis-regulation of specific miRNAs has been demonstrated in a variety of diseases in humans including inflammation and immune disorders. Despite several studies, the multi-factorial mechanisms regulating inflammation is yet unknown. Our long term goal is to investigate the role of miRNAs in the mechanism of regulation of inflammation. We will use constitutively pro-inflammatory Cystic Fibrosis (CF) cells as well as airway smooth muscle cells as appropriate model systems to determine the mechanism of dys-regulated inflammation.
- Moreover, we are also investigating the mechanism by which the processing of miR-155 is regulated in a pro-inflammatory cellular environment using cell culture models, including cystic fibrosis cell lines and mouse macrophages cell lines.
